Golden Gate Assembly

Golden Gate Assembly is a molecular cloning technique that joins multiple DNA fragments in a defined order, enabling efficient construction of plasmids and other genetic designs. It uses Type IIS restriction enzymes, which cut outside their recognition sites, together with DNA ligase in a single reaction; designed four-base overhangs guide fragment assembly, while repeated digestion and ligation selectively enrich the correctly assembled product. Because the recognition sites are removed during assembly, the final construct can be seamless and scarless. The method supports modular cloning, multiplexed DNA assembly, and rapid generation of genetic circuits, expression vectors, and engineered biological systems.
