Growth Plate Analysis

Growth plate analysis is the study of epiphyseal cartilage at the ends of developing long bones, where skeletal lengthening occurs through endochondral ossification. It typically combines imaging or histological examination of the resting, proliferative, hypertrophic, and ossification zones to assess chondrocyte organization, matrix changes, vascular invasion, and bone formation. In biology, this analysis helps characterize normal skeletal development and identify how hormones, genetic changes, disease, drugs, or injury alter growth-plate structure and function, supporting research into bone growth, developmental disorders, and potential regenerative or therapeutic strategies.

This protocol describes an improved mouse model for adolescent bone growth plate injuries. Using transgenic mice with tri-lineage fluorescent reporters for collagen types I, II, and X, the primary matrices associated with three different substrata of the growth plate, injury placement is guided by native fluorescence under the microscope.
