Immobilized Metal Chromatography

Immobilized Metal Chromatography is a protein purification technique that separates molecules according to their affinity for metal ions attached to a solid support. In biological applications, chelating groups immobilize ions such as nickel or cobalt, which coordinate with electron-donating amino acids, especially histidine residues; proteins bind under suitable buffer conditions and are released by adding a competing ligand or changing pH. This selective interaction enables purification of recombinant proteins carrying polyhistidine tags from complex cell lysates through binding, washing, and elution steps. The method supports protein production, structural studies, enzyme research, and downstream biochemical assays.

We present an assay for easy quantification of metals introduced to samples prepared using immobilized metal affinity chromatography. The method uses hydroxynaphthol blue as the colorimetric metal indicator and a UV-Vis spectrophotometer as the detector.

Presented here is a protocol for appending peptide CD47 (pepCD47) to metal stents using polybisphosphonate chemistry. Functionalization of metal stents using pepCD47 prevents the attachment and activation of inflammatory cells thus improving their biocompatibility.
