Immunosuppressive Assay

An immunosuppressive assay is a laboratory test used to measure how effectively a substance, cell population, or treatment reduces immune-system activity. These assays commonly stimulate immune cells, such as lymphocytes, and then quantify changes in cell proliferation, cytokine production, activation markers, or viability compared with untreated controls. By providing measurable evidence of immune suppression, the method supports screening and characterization of immunosuppressive drugs, biologics, and cellular therapies. Immunosuppressive assays also help investigate immune regulation, assess treatment potency, and evaluate potential effects on inflammatory or autoimmune responses in biological research.

Using optimized human peripheral blood mononuclear cell (PBMC) ex vivo assays, we showed that a combustible tobacco product preparation markedly suppresses receptor-mediated intracellularly secreted cytokines and cytolytic ability of effector PBMCs. These rapid assays may be useful in product evaluation and understanding the potential long-term effects of tobacco exposure.
