Latent Tb

Latent TB is a state in which a person is infected with Mycobacterium tuberculosis but has no symptoms and cannot transmit the bacteria through the air, making it an important stage in tuberculosis biology and prevention. It develops when immune cells, including macrophages and lymphocytes, restrict bacterial growth within organized granulomas, although some bacilli can persist in a metabolically quiet state. Tuberculin skin testing and interferon-gamma release assays detect immune responses associated with infection, while clinical evaluation helps exclude active disease. Because immune control can weaken, latent TB may reactivate into contagious disease; identifying and treating infection with preventive therapy reduces this risk and supports tuberculosis control.

Current methods of analyzing patients’ adherence to complex drug resistant-tuberculosis (DR-TB) regimens can be inaccurate and resource-intensive. Our method analyzes hair, an easily collected and stored matrix, for concentrations of 11 DR-TB medications. Using LC-MS/MS, we can determine sub-nanogram drug levels that can be utilized to better understand drug adherence.

We describe a tuberculosis molecular bacterial load assay test performed after heat inactivation of sputum. Heat inactivation renders sputum samples noninfectious and obviates the need for containment level 3 laboratories for tuberculosis molecular tests.

Here, we demonstrate the preparation of Mtb culture filtrate (CF), which is the bacterial growth media that consists of proteins secreted by the bacteria. Mtb secretes proteins to modulate host signaling and immune response, which are crucial for pathogenesis. The CFs prepared are analyzed by mass spectrometry to identify all the secretory factors.

γ-Herpesviruses (γ-HVs) establish life-long persistency in their host. Infection of mice with γ-HV68 provides a genetically tractable in vivo model for the characterization of the lifecycle/pathogenesis of γHVs. This protocol describes the detection and quantitation of γHV68 infection at acute and latent stages following infection by plaque-forming, infectious center, and qPCR assays.
