Mesenchymal Properties

Mesenchymal properties are cellular characteristics associated with a motile, adaptable state that supports tissue remodeling, development, and repair. Cells with these properties typically adopt an elongated, spindle-like shape, reduce stable cell-cell adhesion, reorganize the actin cytoskeleton, and interact dynamically with the extracellular matrix through adhesion structures and matrix-degrading enzymes. In biology, mesenchymal properties are central to processes such as embryonic morphogenesis, wound healing, fibrosis, and cancer invasion, where cells migrate through tissues and respond to biochemical and mechanical signals. Studying these traits helps researchers understand epithelial-mesenchymal transition, tissue organization, and how altered cell behavior contributes to disease.

The immunomodulatory properties of human mesenchymal stem cells (MSC) appear increasingly relevant for clinical application. Using a co-culture system of MSCs and peripheral blood leukocytes pre-stained with the fluorescent dye carboxyfluorescein succinimidyl ester (CFSE), we describe the in vitro assessment of MSC immunomodulation on effector leukocyte proliferation and specific subpopulations.

Electroporation of Craniofacial Mesenchyme

Craniofacial cartilages develop in close contact with other tissues and are difficult to manipulate in live animals. We are using electroporation to deliver molecular tools during growth of the craniofacial skeleton while bypassing early embryonic effects. This approach will allow us to efficiently test candidate molecules in vivo.

We present here a cell culture method for inducing mesenchymal-epithelial transitions (MET) in sarcoma cells based on combined ectopic expression of microRNA-200 family members and grainyhead-like 2 (GRHL2). This method is suitable for better understanding the biological impact of phenotypic plasticity on cancer aggressiveness and treatments.
