Microbial Culture Fitness

Microbial culture fitness describes how effectively microorganisms grow, survive, and reproduce under defined laboratory conditions, making it a useful measure of population performance. It reflects the balance between nutrient use, cellular stress responses, metabolic activity, and competition among cells as environmental conditions shape growth rate and biomass production. Measuring culture fitness can reveal how microbes respond to changes in temperature, pH, oxygen availability, nutrient supply, or antimicrobial exposure. In biology, these assessments support strain selection, experimental reproducibility, microbial ecology, and studies of adaptation, helping researchers link cellular traits to population-level outcomes.

Quantitative Fitness Analysis (QFA) is a complementary series of experimental and computational methods for estimating microbial culture fitnesses. QFA estimates the effect of genetic mutations, drugs or other applied treatments on microbe growth. Experiments scaling from focussed analysis of single cultures to thousands of parallel cultures can be designed.

Source: Jian, X., et al., Automated Microbial Cultivation and Adaptive Evolution using Microbial Microdroplet Culture System (MMC). J. Vis. Exp. (2022).This video demonstrates the use of a microbial microdroplet culture system to drive adaptive evolution of bacteria toward methanol tolerance. By mixing engineered bacterial cultures with a high-methanol stress medium and segmenting them into microdroplets, the system selects for mutants that exhibit improved growth under methanol stress. Through...
