Microcentrifuge Tube Incubation

Microcentrifuge tube incubation is the controlled holding of small-volume biological samples or reaction mixtures in sealed tubes at a defined temperature and for a set period. During incubation, heat transfer from a water bath, heat block, or similar device promotes processes such as enzymatic reactions, cell lysis, nucleic acid preparation, or molecular binding while the tube contains the sample and limits contamination. This routine laboratory method supports DNA and RNA workflows, protein analysis, microbiology, and biochemical assays. Careful control of temperature, duration, mixing, and tube closure improves reproducibility and helps protect samples from evaporation or degradation.

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.
