Microrna Quantitation

MicroRNA quantitation is the measurement of small, noncoding RNA molecules that regulate gene expression, providing insight into cellular state and biological function. In common workflows, researchers extract RNA from cells or tissues, convert microRNAs into complementary DNA through reverse transcription, and amplify specific sequences using quantitative PCR, with reference controls supporting relative or absolute measurement. This approach helps characterize developmental programs, stress responses, disease-associated expression changes, and regulatory networks. Accurate quantitation can reveal biomarkers, clarify post-transcriptional control, and support studies of how microRNA profiles change across tissues, experimental conditions, or stages of biological processes.

Cerebrospinal Fluid MicroRNA Profiling Using Quantitative Real Time PCR

We describe a protocol of real time PCR to profile microRNAs in the cerebrospinal fluid (CSF). With the exception of RNA extraction protocols, the procedure can be extended to RNA extracted from other body fluids, cultured cells, or tissue specimens.

Quantitative Real Time polymerase chain reaction (qPCR) is a rapid and sensitive method to investigate the expression levels of various microRNA (miRNA) molecules in tumor samples. Using this method expression of hundreds of different miRNA molecules can be amplified, quantified, and analyzed from the same cDNA template.

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns—non-coding regions of a gene—or intergenic regions—stretches of DNA present between genes. We will demonstrate the setup and analysis of pre-microRNA 96-well arrays for QPCR using a robot as well as by hand with a Thermo Scientific Matrix multichannel pipette.
