Mycelial Pellet Sorting

Mycelial pellet sorting is the separation and classification of compact fungal cell aggregates formed during submerged culture, a process that helps standardize samples for biological analysis and production. It works by isolating pellets from the surrounding liquid and grouping them according to measurable features such as size, shape, or density, using approaches that may include sieving, filtration, or image-based analysis. In biology and fungal biotechnology, sorting supports consistent inoculum preparation, improves comparisons of growth and morphology, and helps evaluate how culture conditions affect pellet formation. These measurements can guide fermentation optimization and improve the reproducibility of downstream experiments.

Liquid-grown Streptomyces cultures are characterized by mycelial pellets that are heterogeneous in size. We here describe a method to analyze and sort such pellets in a high-throughput manner. These pellets can be used for further analyses, which will provide leads to understand and control growth heterogeneity.

Microplastics act as vector of potentially toxic organic contaminants with unpredictable effects. This protocol describes an alternative methodology for assessing the levels of organochlorine pesticides adsorbed on plastic pellets and identifying the polymer chemical structure. The focus is on pressurized fluid extraction and attenuated total reflectance Fourier transform infrared spectroscopy.

Method to Produce Durable Pellets at Lower Energy Consumption Using High Moisture Corn Stover and a Corn Starch Binder in a Flat Die Pellet Mill

In this study, a protocol was developed to produce good quality pellets using a flat die pellet mill at reduced specific energy consumption testing high-moisture corn stover and a starch based binder. The results indicated that adding a corn starch binder improved the pellet durability, reduced percent fines and decreased specific energy consumption.

Fluorescence Activated Cell Sorting of Plant Protoplasts

A method for isolating specific cell types from plant material is demonstrated. This technique employs transgenic marker lines expressing fluorescent proteins in particular cell types, cellular dissociation and Fluorescence Activated Cell Sorting. Additionally, a growth setup is established here that facilitates treatment of Arabidopsis thaliana seedlings prior to cell sorting.
