Neural Tracing Technique

Neural tracing technique is a group of methods used to map connections between neurons and brain regions, revealing how nervous systems are organized. Tracers are introduced into a selected site and transported along axons in anterograde direction toward synaptic targets or retrograde direction toward cell bodies, where microscopy identifies labeled pathways. Fluorescent dyes, genetically encoded markers, or viral vectors can provide structural and sometimes cell-type-specific information. In biology, neural tracing supports circuit mapping, studies of sensory and motor systems, and investigation of changes associated with development, injury, or neurological disease.

Capitalizing on a binary genetic strategy we provide a detailed protocol for neural circuit tracing in mice that express complementary transsynaptic tracers after Cre-mediated recombination. Because cell-specific tracer production is genetically encoded, our experimental approach is suitable to study the formation and maturation of neural circuitry during murine embryonic brain development at a single cell resolution.
