Patch Clamp Technique

The patch clamp technique is an electrophysiological method for measuring ionic currents across cell membranes, making it essential for studying how cells communicate and respond to stimuli. A glass micropipette forms a tight, high-resistance seal with a small area of membrane, allowing researchers to control membrane voltage or current while recording ion channel activity; configurations such as whole-cell and single-channel recording provide different levels of access. In biology, patch clamp experiments reveal channel conductance, gating, and membrane excitability in neurons, muscle cells, and other tissues. These measurements support research on signaling, pharmacology, disease mechanisms, and therapeutic drug development.

Neuron cell membranes are populated with ion channels that control the movement of charge into and out of the cell, thereby regulating neuron firing. One extremely useful technique for investigating the biophysical properties of these channels is called patch clamp recording. In this method, neuroscientists place a polished glass micropipette against a cell and apply suction to form a high resistance seal. This process isolates a small “patch” of membrane that contains one or more ion channels.
