Rat Heart Surgery

Rat heart surgery is a set of microsurgical procedures used to access, manipulate, or repair the heart in living rats, providing controlled models for investigating cardiovascular biology and disease. Under anesthesia, researchers perform a thoracotomy to expose the heart, maintain physiological conditions, and carry out procedures such as coronary artery manipulation or tissue sampling before carefully closing the chest. These models help characterize cardiac injury, remodeling, ischemia, and recovery while enabling evaluation of potential therapies and surgical techniques. Because rat hearts share key structural and functional features with mammalian cardiovascular systems, the approach remains valuable for translational research, although findings require confirmation in larger models and clinical studies.

This protocol describes a method to collect cardiac interstitial fluid from the isolated, perfused rat heart. To physically separate interstitial transudate from coronary venous effluent perfusate, the Langendorff perfused heart is inverted, and the transudate (interstitial fluid) formed on the cardiac surface is collected using a soft latex cap.
