Rna Chromatin Complex

An RNA chromatin complex is an assembly of RNA molecules, chromatin, and associated proteins that influences how genetic information is organized and expressed in the nucleus. These complexes form when regulatory RNAs bind chromatin directly or interact with DNA-binding proteins, helping recruit chromatin-modifying enzymes that alter nucleosome arrangement and control access to genes. By linking RNA activity with epigenetic regulation, they contribute to transcriptional silencing, gene activation, dosage compensation, and genome stability. Studying RNA chromatin complexes helps clarify gene regulation in development and disease and supports research into therapies that target abnormal epigenetic states.

Sequential salt extraction of chromatin bound proteins is a useful tool for determining the binding properties of large protein complexes. This method can be employed to evaluate the role of individual subunits or domains in the overall affinity of a protein complex to bulk chromatin.

Influenza viruses replicate their RNA genome in association with host-cell chromatin. Here, we present a method to purify intact viral ribonucleoprotein complexes from the chromatin of infected cells. Purified viral complexes can be analyzed by both Western blot and primer extension of protein and RNA content, respectively.

Generation and Purification of Human INO80 Chromatin Remodeling Complexes and Subcomplexes

This protocol describes a procedure for generating and purifying wild type and mutant versions of the human INO80 chromatin remodeling complex. Epitope tagged versions of INO80 subunits are stably expressed in HEK293 cells, and complete complexes and complexes lacking specific sets of subunits are purified by immunoaffinity chromatography.
