Stem Cell Pool

A stem cell pool is a population of stem cells maintained within a tissue to support growth, repair, and ongoing cell replacement. Through asymmetric or symmetric division, these cells balance self-renewal, which preserves the pool, with differentiation, which produces specialized progeny; signals from the surrounding stem cell niche help regulate this balance. In biology, studying stem cell pools clarifies how tissues develop, maintain homeostasis, and respond to injury. This knowledge supports research into regenerative medicine, disease mechanisms, aging, and cancer, where altered stem cell behavior can contribute to impaired repair or uncontrolled growth.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Mouse Epidermal Neural Crest Stem Cell (EPI-NCSC) Cultures

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

MISSION LentiPlex Pooled shRNA Library Screening in Mammalian Cells

Here we use a human LentiPlex pooled library and traditional sequencing methods to identify gene targets promoting cell survival. We demonstrate how to set up and deconvolute a LentiPlex screen and validate the results.
