T Cell Development

T cell development is the process by which immature lymphoid cells become functional T lymphocytes, a central feature of adaptive immunity. Precursors migrate to the thymus, where they rearrange T cell receptor genes and progress through defined stages marked by CD4 and CD8 expression; positive selection preserves cells that recognize self major histocompatibility complex molecules, while negative selection removes strongly self-reactive cells. This coordinated maturation establishes a diverse yet self-tolerant T cell repertoire. Understanding T cell development supports research into immune deficiencies, autoimmunity, thymic biology, cancer immunology, and strategies for engineering or restoring immune function.

Single-cell Profiling of Developing and Mature Retinal Neurons

A method for the isolation of single retinal cells and subsequent amplification of their cDNAs is described. Single-cell transcriptomics reveals the degree of cellular heterogeneity present in a tissue and uncovers new marker genes for rare cell populations. The accompanying protocol can be adjusted to suit many different cell types.

High-resolution Live Imaging of Cell Behavior in the Developing Neuroepithelium

Imaging embryonic tissue in real-time is challenging over long periods of time. Here we present an assay for monitoring cellular and sub-cellular changes in chick spinal cord for long periods with high spatial and temporal resolution. This technique can be adapted for other regions of the nervous system and developing embryo.

Live Imaging of Cell Extrusion from the Epidermis of Developing Zebrafish

Dying cells are extruded from epithelial tissues by concerted contraction of neighboring cells without disrupting barrier function. The optical clarity of developing zebrafish provides an excellent system to visualize extrusion in living epithelia. Here we describe methods to induce and image extrusion in the larval zebrafish epidermis at cellular resolution.

Here, we present a method for the photoactivated switch of photoconvertible fluorescent proteins (PCFPs) in the living zebrafish embryo and further tracking of photoconverted protein at specific time points during development. This methodology allows monitoring of cell biological events underlying different developmental processes in a live vertebrate organism.
