Collagen Matrix Invasion

Collagen matrix invasion is the movement of cells, particularly cancer cells, through a three-dimensional collagen network that models the extracellular matrix surrounding tumors. In this process, cells attach to collagen through adhesion receptors, generate contractile forces, and may secrete matrix-degrading enzymes that create or enlarge paths through the matrix; cell shape, matrix density, and stiffness influence migration. In cancer research, collagen invasion assays provide controlled models for measuring invasive behavior and examining how tumor cells interact with their physical environment. These models help evaluate mechanisms of local invasion, compare metastatic potential, and assess compounds that alter cell motility or extracellular-matrix remodeling.

In order to understand the mechanisms that can alter cell migration, we present a planar gradient diffusion system to investigate chemotaxis in a 3D collagen matrix, which allows one to overcome modern diffusion chamber limitations of existing assays.

Explants from the midbrain dopamine system and striatum are used in a collagen matrix assay for the in vitro analysis of mesostriatal and striatonigral pathway development. In this assay axonal outgrowth and guidance can be manipulated and quantified. It can also be modified for assessing other regions or molecular cues.

In this video, glioblastoma and microglia cells are mixed with a polymer matrix solution and transferred to a porous chamber insert where a gel forms, embedding the cells.
