Mucosal Separation

Mucosal separation is a tissue-processing method that isolates the mucosal layer from deeper wall structures, enabling focused study of epithelial and stromal compartments in cancer research. The approach uses careful mechanical dissection or layer-specific processing to separate the mucosa from underlying submucosa and muscle while preserving material for downstream analysis. Researchers can then examine tissue morphology, gene expression, proteins, or cellular changes within the mucosa and compare them with adjacent layers or normal controls. This compartment-specific analysis helps clarify how tumors arise, interact with their local microenvironment, and respond to treatment, supporting more precise investigation of gastrointestinal and other mucosa-associated cancers.

A mouse model of human endoscopic skull base reconstruction has been developed that creates a semipermeable interface between the brain and nose using nasal mucosal grafts. This method allows researchers to study delivery to the central nervous system of high molecular weight therapeutics which are otherwise excluded by the blood-brain barrier when administered systemically.

This manuscript describes the production, characterization and potential uses of a tissue engineered 3D esophageal construct prepared from normal primary human esophageal fibroblast and squamous epithelial cells seeded within a de-cellularized porcine scaffold. The results demonstrate the formation of a mature stratified epithelium similar to the normal human esophagus.
