Suspension Cell Analysis

Suspension cell analysis is the examination of cells dispersed in a liquid medium to measure their physical, chemical, and biological properties. In practice, representative samples are assessed using techniques such as optical imaging, particle counting, or flow-based measurements, often with fluorescent stains that distinguish viable cells, dead cells, or specific molecular features. These analyses help monitor cell concentration, size, morphology, viability, and biochemical responses during culture. In chemistry and bioprocess research, suspension cell analysis supports optimization of growth conditions, evaluation of drug or reagent effects, quality control, and investigation of how cells respond to changes in their chemical environment.

This article describes spectral cytometry, a new approach in flow cytometry that uses the shapes of emission spectra to distinguish fluorochromes. An algorithm replaces compensations and can treat auto-fluorescence as an independent parameter. This new approach allows for the proper analysis of cells isolated from solid organs.

The skin is home to a complex immune cell network. We describe an efficient methodology for the digestion of mouse skin, from different parts of the animal's body, in order to obtain a single-cell suspension and analyze the different leukocyte populations resident in the skin by flow cytometry.
