Truncated Sgrna

Truncated single-guide RNA (sgRNA) is a shortened CRISPR guide molecule designed to direct Cas9 to a selected DNA sequence while limiting unintended targeting. It retains the scaffold that binds Cas9 but uses a shorter target-complementary spacer, commonly 17–18 nucleotides rather than the conventional 20; reduced guide-DNA pairing can make Cas9 less tolerant of mismatches at off-target sites, although editing activity may also decrease. In genetics, truncated sgRNAs support more selective genome editing for gene disruption, sequence modification, and functional studies, where balancing on-target efficiency against off-target effects is essential. Researchers evaluate their performance by measuring intended edits and unintended genomic changes.

Truncation in survival analysis refers to the exclusion of individuals or events from the dataset based on specific criteria related to the time of the event. This exclusion can happen in two primary forms: left truncation and right truncation. Left truncation occurs when individuals who experienced the event of interest before a certain time are not included in the study. This is often due to a "delayed entry" into the study where only those who survive until a certain entry point are observed.

This video demonstrates a technique for nanoblade-based delivery of Cas9-sgRNA ribonucleoprotein complex inside target cells for genome editing. Nanoblades are virus-like particles devoid of the ability to multiply and infect neighboring cells, therefore are very useful for rapid and dose-dependent transportation of bio- and nanomaterials.

A CRISPR/sgRNA library has been applied to interrogating protein-coding genes.However, the feasibility of a sgRNA library to uncover the function of a CTCF boundary in gene regulation remains unexplored. Here, we describe a HOX loci specific sgRNA library to elucidate the function of CTCF boundaries in HOX loci.

We have developed a simple and inexpensive protocol to load Cas9/single-guide RNA (sgRNA) ribonucleoprotein complexes within virus-like particles. These particles, called "Nanoblades", allow efficient delivery of the Cas9/sgRNA complex in immortalized and primary cells as well as in vivo.
