Antibiotic Cocktail Preparation

Antibiotic cocktail preparation is the controlled formulation of two or more antimicrobial agents into a working solution, enabling coordinated treatment or experimental selection against microbial infection. Preparation requires choosing compatible antibiotics, calculating target concentrations, dissolving or diluting each agent appropriately, and maintaining sterile conditions while accounting for stability, solvent compatibility, and storage. In immunology and infection research, antibiotic cocktails help suppress contaminating bacteria, select genetically modified microorganisms, or model combination therapy, while careful formulation supports reproducible dosing and reduces errors caused by degradation or incompatible components. Their use can also help investigators assess antimicrobial interactions and treatment outcomes.

Whole blood Immunophenotyping is indispensable for monitoring the human immune system. However, the number of reagents required and the instability of specific fluorochromes in premixed cocktails necessitates daily reagent preparation. Here we show that semi-automated preparation of staining antibody cocktail helps establish reliable immunophenotyping by minimizing variability in reagent dispensing.

Multi-exon Skipping Using Cocktail Antisense Oligonucleotides in the Canine X-linked Muscular Dystrophy

Exon skipping is currently a most promising therapeutic option for Duchenne muscular dystrophy (DMD). To expand the applicability for DMD patients and to optimize the stability/function of the resulting truncated dystrophin proteins, a multi-exon skipping approach using cocktail antisense oligonucleotides was developed and we demonstrated systemic dystrophin rescue in a dog model.

Antibiotic Dereplication Using the Antibiotic Resistance Platform

We describe a platform that utilizes a library of isogenic antibiotic resistant Escherichia coli for the dereplication of antibiotics. The identity of an antibiotic produced by bacteria or fungi can be deduced by the growth of E. coli expressing its respective resistance gene. This platform is economically effective and time-efficient.
