Cell Morphology Tracing

Cell morphology tracing is a microscopy-based method for documenting and analyzing the shape, boundaries, and structural changes of individual cells. It works by identifying cellular features in images and following their outlines or positions across samples or time points, allowing researchers to quantify changes in size, form, and spatial organization. In immunology and infection research, morphology tracing helps characterize immune-cell activation, migration, adhesion, and interactions with pathogens or infected cells. These measurements can reveal how cellular structure changes during host defense or disease progression, supporting comparisons between experimental conditions and improving the interpretation of cellular responses.

Automated Sholl Analysis of Digitized Neuronal Morphology at Multiple Scales

We have developed a computer program to analyze neuronal morphology. In combination with two existing open source analysis tools, our program performs Sholl analysis and determines the number of neurites, branch points, and neurite tips. The analyses are performed so that local changes in neurite morphology can be observed.

In this article we describe the principles for designing and performing a multi-color lineage tracing experiment using R26R-Confetti mice. We provide a specific protocol for tracking corneal epithelial cells which can be modified for other tissues of interest.
