Neutralization Sensitivity

Neutralization sensitivity is the degree to which a pathogen, typically a virus, can be blocked by antibodies or other neutralizing factors, making it an important measure of immune protection. It depends on how effectively antibodies bind accessible epitopes and interfere with essential steps such as receptor attachment, membrane fusion, or cellular entry. Measuring neutralization sensitivity helps compare viral strains, evaluate vaccine- and infection-induced immunity, and assess therapeutic antibody activity. Changes in viral surface proteins can alter antibody recognition, providing insight into immune escape, variant emergence, and the durability of protection in immunology and infection research.

Optimization of a Quantitative Micro-neutralization Assay

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

Here, we present a protocol to detect neutralization epitopes on antigen-displaying virus-like particles (VLPs). Immunoprecipitation of the human immunodeficiency virus (HIV)-derived VLPs is performed using envelope glycoproteins-specific monoclonal antibodies coupled to protein G-conjugated magnetic beads. Captured VLPs are subsequently subjected to SDS-PAGE and Western blot-analysis employing viral core protein Gag-specific antibodies.

Generation of Influenza Virus Escape Variants against Neutralizing Antibodies

This video demonstrates a technique for generating escape variants of neutralizing influenza virus monoclonal antibodies. Antibody-neutralized virus suspension is injected into embryonated chicken eggs to propagate escape variants selectively. This results in a virus population predominantly of escape variants, confirmed via a Hemagglutination assay.
