Fiber Structural Analysis

Fiber structural analysis is the systematic characterization of a fiber’s organization, dimensions, composition, and physical properties, helping researchers relate microscopic architecture to biological function and clinical performance. It typically combines microscopy or other imaging with measurements of fiber diameter, alignment, packing, orientation, and surface features; these observations can reveal how processing, disease, or injury changes structure. In medicine, the approach supports evaluation of collagen-rich tissues, nerve and muscle fibers, biomaterial scaffolds, and fibrous medical devices, providing evidence for diagnosis, tissue repair, implant design, and quality control. Standardized structural measurements also improve comparison across samples and guide development of safer, more effective therapies.

Correlative Microscopy for 3D Structural Analysis of Dynamic Interactions

We describe a correlative microscopy method that combines high-speed 3D live-cell fluorescent light microscopy and high-resolution cryo-electron tomography. We demonstrate the capability of the correlative method by imaging dynamic, small HIV-1 particles interacting with host HeLa cells.

We describe a novel approach for simultaneous analysis of brain function and structure using magnetic resonance imaging (MRI). We assess brain structure with high-resolution diffusion-weighted imaging and white-matter fiber tractography. Unlike standard structural MRI, these techniques allow us to directly relate anatomical connectivity to functional properties of brain networks.

This protocol uses three-dimensional (3D) imaging and analysis techniques to visualize and quantify nerve-specific mitochondria. The techniques are applicable to other situations where one fluorescent signal is used to isolate a subset of data from another fluorescent signal.
