Lung Mechanics

Lung mechanics is the study of how pressure, airflow, volume, and tissue forces govern ventilation, providing a foundation for understanding normal breathing and respiratory disease. During inspiration, contraction of the diaphragm lowers pleural pressure, creating a transpulmonary pressure gradient that expands the lungs; compliance reflects how readily they stretch, while airway resistance influences airflow, especially during expiration. Measuring pressure-volume relationships, respiratory system compliance, and resistance helps clinicians interpret conditions such as asthma, emphysema, and pulmonary fibrosis. These principles also guide mechanical ventilation by supporting appropriate airway pressures and reducing risks associated with overdistension or inadequate lung expansion.

The stiffness of the extracellular matrix strongly influences multiple behaviors of adherent cells. Matrix stiffness varies spatially throughout a tissue, and undergoes modification in various disease conditions. Here we develop methods to characterize spatial variations in stiffness in normal and fibrotic mouse lung tissue using atomic force microscopy microindentation.

Early embryonic lung organ culture is a very useful system to study epithelial-mesenchymal interactions. Both epithelial and mesenchymal morphogenesis proceeds under specific conditions that can be readily manipulated in this system.

A Simple Method of Mouse Lung Intubation

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.

Ex-Vivo Lung Perfusion (EVLP) has allowed lung transplantation in humans to become more readily available by enabling the ability to assess organs and expand the donor pool. Here, we describe the development of a rat EVLP program and refinements that allow for a reproducible model for future expansion.
