Worm Tracking Software

Worm tracking software is a computer-vision tool that detects, follows, and quantifies the movement of nematodes in recorded images or video. It typically segments each worm from the background, identifies body position or a central point, and links these measurements across frames to generate trajectories and behavioral metrics such as speed, distance traveled, and turning frequency. In bioengineering, these analyses support standardized studies of locomotion, neuromuscular function, environmental responses, and phenotypic effects of genetic or chemical perturbations. Automated tracking also improves experimental throughput and reproducibility by reducing manual scoring and enabling quantitative comparisons across individuals and conditions.

Screening for mutants with phenotypic defects is a straightforward method for identifying genes that function in a given biological process. In this article we describe how to culture free living worms (e.g., Pristionchus pacificus) in the laboratory and show two different mutagenesis methods, EMS and TMP/UV.

Locomotor activity (LMA) is a simple and easily performed measurement of behavior in mice. Coupling of video tracking software (VTS) and LMA allows for the improvement of specificity and sensitivity, especially when compared with the manual, line crossing method of LMA analysis. Additionally VTS allows long-term tracking of mouse LMA.

The current study aimed to automate the quantification of motor deficits in rats. The initial evaluation model assesses motor loss resulting from an intracortical microelectrode implantation in the motor cortex. We report on the development and use of a tracking algorithm using easily adaptable, simple, and readily available coding software.
