Serum Free Medium

Serum-free medium is a cell-culture formulation that supports the growth, survival, or differentiation of cells without adding animal serum, making experimental conditions more defined and reproducible. Instead of relying on the complex mixture of proteins, lipids, hormones, and growth factors in serum, it supplies selected nutrients and supplements, often including defined growth factors, trace elements, and attachment components tailored to a cell type. In biology research, these media reduce batch-to-batch variation and unwanted biological signals, improving the interpretation of cell responses. They are used in primary cell culture, stem cell studies, vaccine production, and biomanufacturing when controlled conditions are essential.

Efforts to understand microglial function in detail have been hindered by the lack of microglial culture models that recapitulate the properties of mature in vivo microglia. This protocol describes an isolation and culture approach designed to maintain robust survival of highly ramified mature rat microglia under defined-medium conditions.
