Retrospective Analysis

Retrospective analysis is a research method that examines existing records or previously collected data to identify patterns, associations, and outcomes over time. In cancer research, investigators define a study population, extract clinical, pathological, treatment, or survival information from medical records and databases, and analyze relationships among exposures, interventions, and outcomes without assigning treatments prospectively. This approach supports evaluation of treatment effectiveness, prognostic factors, disease progression, and patient characteristics in real-world settings. Although retrospective studies can be efficient and valuable for generating hypotheses, researchers must address incomplete records, selection bias, confounding variables, and differences in follow-up when interpreting findings.

Salmonella invades and replicates inside intestinal epithelial cells both in Salmonella-specific vacuoles and free in the cytosol (hyper-replication). A high-throughput fluorescence microscopy-based protocol is described here to quantify the intracellular phenotypes of Salmonella by two complementary image analyses through ImageJ, reaching single-cell resolution and scoring.
