Subcellular Structure Dynamics

Subcellular structure dynamics is the study of how organelles, cytoskeletal elements, membranes, and molecular assemblies change their organization, position, and interactions over time. These changes arise through processes such as protein assembly and disassembly, membrane remodeling, motor-protein transport, and regulated signaling, allowing cells to adapt their internal architecture to developmental cues. In developmental biology, dynamic control of subcellular structures helps establish cell polarity, coordinate division, guide migration, and support differentiation as tissues form. Imaging, live-cell analysis, and molecular perturbation can reveal how disruptions in these dynamics contribute to abnormal development and disease.

Imaging Subcellular Structures in the Living Zebrafish Embryo

Imaging the dynamic behavior of organelles and other subcellular structures in vivo can shed light on their function in physiological and disease conditions. Here, we present methods for genetically tagging two organelles, centrosomes and mitochondria, and imaging their dynamics in living zebrafish embryos using wide-field and confocal microscopy.

Imaging-derived mean square displacement (iMSD) analysis is applied to macropinosomes to highlight their intrinsic time-evolving nature in terms of structural and dynamic properties. Macropinosomes are then compared to insulin secretory granules (ISGs) as a reference for subcellular structures with time-invariant average structural/dynamic properties.
