Capture Antibody Binding

Capture antibody binding is an immunoassay principle in which an immobilized antibody selectively retains a target antigen from a biological sample, enabling its detection and measurement. The capture antibody recognizes a specific epitope through complementary molecular interactions, while a second, labeled detection antibody can bind another epitope to form a sandwich complex; the resulting signal reflects the amount of captured antigen. In immunology and infection research, this approach supports sensitive detection of pathogen components, antibodies, or immune mediators in specimens such as serum and culture fluids. It is widely used in diagnostic testing, biomarker analysis, and evaluation of host responses to infection.

Conversion of a Capture ELISA to a Luminex xMAP Assay using a Multiplex Antibody Screening Method

An ELISA can be easily converted to a Luminex xMAP assay and, through the benefits of multiplexing, several antibodies can be screened simultaneously to identify an optimum antibody pair, resulting in increased sensitivity and dynamic range, while reducing assay cost.

Capture Compounds are trifunctional small molecules to reduce the complexity of the proteome by functional reversible small molecule-protein interaction followed by photo-crosslinking and purification. Here we use a Capture Compound with S-adenosyl-L-homocysteine-binding as selectivity function to isolate methyltransferases from an Escherichia coli whole cell lysate and identify them by MS.

This video describes the use of low-magnification, dark-field microscopy to quantify specific exosomes in small volume biofluidic samples. The exosomes thus identified can serve as potential cancer biomarkers.
