Neural Rosette Stage

The neural rosette stage is an early pattern of neural development in which neural progenitor cells organize into radial structures that resemble the embryonic neural tube. During this stage, polarized progenitors form apical junctions around a central lumen, establishing coordinated cell architecture and supporting neural fate specification. Neural rosettes commonly arise during the differentiation of pluripotent stem cells and provide an accessible model of early neurogenesis. Researchers use them to study neural patterning, progenitor behavior, and developmental disorders, as well as to evaluate how genetic changes or treatments affect the formation and organization of human neural tissue.

We have developed an in vitro unfolded hippocampus which preserves CA1-CA3 array of neurons. Combined with the penetrating micro-electrode array, neural activity can be monitored in both the longitudinal and transverse orientations. This method provides advantages over hippocampal slice preparations as the propagation in the entire hippocampus can be recorded simultaneously.

Chick in ovo electroporation is a technique which allows genetic manipulation of the avian embryo. Common applications of this technique include functional analysis of genes and putative enhancer elements. This video demonstrates neural tube electroporation in HH 10 chick embryos. Injection technique and proper egg handling are discussed.
