Retrograde Labeling

Retrograde labeling is a neuroscience tracing method that identifies neurons projecting to a selected target region by marking them from their axon terminals. Researchers introduce a tracer into the target tissue, where it is taken up by nerve endings and transported backward along the axon to the cell body, typically through microtubule-based transport. Afterward, labeled neurons can be visualized with microscopy and analyzed by their location, morphology, or molecular characteristics. This approach helps map anatomical connections, define neural pathways, compare projection patterns across conditions, and relate circuit organization to behavior, development, or neurological disease.

Retrograde transport of fluorescent dye labels a sub-population of neurons based on anatomical projection. Labeled axons can be visually targeted in vivo, permitting extracellular recording from identified axons. This technique facilitates recording when neurons cannot be labeled through genetic manipulation or are difficult to isolate using 'blind' in vivo approaches.
