Adventitia

The adventitia is the outer connective-tissue layer surrounding many tubular organs and blood vessels, where it provides structural support and links them to nearby tissues. Composed primarily of collagen and elastic fibers, it anchors vessels and organs, while its nerves, lymphatic vessels, and, in large arteries and veins, vasa vasorum help regulate and nourish the surrounding wall. In biology, studying the adventitia clarifies how tissues maintain mechanical stability, communicate with neighboring structures, and respond to injury. Its remodeling contributes to vascular inflammation, fibrosis, and diseases such as atherosclerosis and aneurysm, making it relevant to histology, pathology, and cardiovascular research.

This paper presents a flow cytometry-based method to investigate the immune composition of aortas. The paper also illustrates an additional technique that allows examining surrounding adventitia and vessel wall separately. This method opens possibilities to perform phenotypical analyses of aortic leukocytes and apply several immunological assays for atherosclerosis studies.

Preparing Cerebral Endothelial Tubes from Mouse Parenchymal Arterioles

This is a guideline for constructing in vivo vascularized tissue using a microsurgical arteriovenous loop or a flow-through pedicle configuration inside a tissue engineering chamber. The vascularized tissues generated can be employed for organ regeneration and replacement of tissue defects, as well as for drug testing and disease modeling.

2013

We present a preparation for visualizing and manipulating calcium signaling in native, intact microvascular endothelium. Endothelial tubes freshly isolated from mouse resistance arteries supplying skeletal muscle retain in vivo morphology and dynamic signaling within and between neighboring cells. Endothelial tubes can be prepared from microvessels of other tissues and organs.

Simultaneous Measurement of Intracellular Calcium and Membrane Potential in a Mouse Cerebral Endothelium

2025

The video demonstrates the simultaneous measurement of intracellular calcium levels and changes in membrane potential in endothelial cells of mouse cerebral artery segments. The isolated endothelial tube is loaded with a fluorescent calcium indicator and treated with a drug that induces intracellular calcium release, resulting in a change in membrane potential. The indicator's fluorescence enables the measurement of calcium levels, and a recording electrode is inserted into a cell to measure...
