Chloroform Extraction

Chloroform extraction is a solvent-based technique that separates biological molecules by exploiting their different solubilities between aqueous and organic phases. In nucleic acid preparation, chloroform is commonly combined with phenol or other reagents, then mixed with a biological sample and centrifuged to produce distinct layers; proteins, lipids, and cellular debris partition into the organic phase or interface, while nucleic acids remain primarily in the aqueous phase. The aqueous fraction can then be recovered for DNA or RNA purification and downstream analysis, including amplification, sequencing, and gene expression studies. Careful handling is essential because chloroform is volatile and hazardous.
