Ecotropic Virus

An ecotropic virus is a virus, typically an enveloped retrovirus, that preferentially infects cells from the species in which it naturally occurs, making host range a defining feature of its biology. In common murine ecotropic viruses, envelope proteins bind the mCAT-1 receptor on mouse cells, allowing membrane entry; the viral RNA is then reverse-transcribed into DNA and integrated into the host genome. This species-restricted tropism helps researchers study viral entry, receptor specificity, replication, and host barriers. Ecotropic retroviral systems are also used in laboratory models and gene-transfer research, where controlled infection of compatible cells can support stable expression of introduced genetic material.

Lentiviruses are a valuable research tool for exploring gene function; however, researchers may wish to avoid production of pantropic lentivirus encoding known or suspected oncogenes. As an alternative, we present a safer protocol for use of ecotropic lentivirus on human cells modified to express the ecotropic receptor mSlc7a1.
