Laser Scanning Confocal Microscopy

Laser scanning confocal microscopy is an optical imaging technique that produces high-resolution, depth-resolved images of fluorescently labeled specimens, making it valuable for examining biological structures in three dimensions. A focused laser beam scans the sample point by point, while a detector positioned behind a pinhole rejects out-of-focus light and collects signal from a defined focal plane. By acquiring sequential optical sections and combining them into a three-dimensional reconstruction, researchers can visualize cells, tissues, organelles, and molecular localization with improved contrast. The method supports studies of cell organization, dynamic biological processes, tissue architecture, and interactions within complex specimens.

Oral Biofilm Analysis of Palatal Expanders by Fluorescence In-Situ Hybridization and Confocal Laser Scanning Microscopy

We present a protocol for structural and compositional analysis of natural oral biofilm from orthodontic appliances with in situ hybridization (FISH) and confocal laser scanning microscopy (CLSM). Oral biofilm samples were collected from palatal expanders, scraping acrylic-resin flakes off their surface and referring them for molecular processing.

The complete construction of a custom, real-time confocal scanning imaging system is described. This system, which can be readily used for video-rate microscopy and microendoscopy, allows for an array of imaging geometries and applications not accessible using standard commercial confocal systems, at a fraction of the cost.

Near Simultaneous Laser Scanning Confocal and Atomic Force Microscopy (Conpokal) on Live Cells

Presented here is the protocol of the Conpokal technique, which combines confocal and atomic force microscopy into a single instrument platform. Conpokal provides same cell, same region, near simultaneous confocal imaging and mechanical characterization of live biological samples.
