Nanoparticle Preparation

Nanoparticle preparation is the design and production of particles with dimensions typically measured on the nanometer scale, where size, shape, composition, and surface properties strongly influence biological behavior. Researchers use top-down methods to break larger materials into nanoscale structures or bottom-up methods in which atoms, molecules, or polymers assemble through nucleation and growth; reaction conditions such as concentration, temperature, pH, and mixing help control particle formation. In biology, prepared nanoparticles can encapsulate drugs, nucleic acids, or imaging agents and be modified with targeting molecules. Reliable preparation supports studies of cellular uptake, biodistribution, diagnostics, therapeutics, and tissue engineering.

Preparation of Silica Nanoparticles Through Microwave-assisted Acid-catalysis

Silica nanoparticles were prepared using acid-catalysis of a siloxane precursor and microwave-assisted synthetic techniques resulting in the controlled growth of nanomaterials ranging from 30-250 nm in diameter. The growth dynamics can be controlled by varying the initial silicic acid concentration, time of the reaction, and temperature of reaction.

Preparation and Characterization of SDF-1α-Chitosan-Dextran Sulfate Nanoparticles

The objective of this protocol is to incorporate SDF-1α, a stem cell homing factor, into dextran sulfate-chitosan nanoparticles. The resultant particles are measured for their size and zeta potential, as well as the content, activity, and in vitro release rate of SDF-1α from the nanoparticles.

Simple homogenization was used to prepare novel, high-density, lipoprotein-mimicking nanoparticles to encapsulate nerve growth factor. Challenges, detailed protocols for nanoparticle preparation, in vitro characterization, and in vivo studies are described in this article.

A number of different procedures for preparing nanoparticles for surface analysis are presented (drop casting, spin coating, deposition from powders, and cryofixation). We discuss the challenges, opportunities, and possible applications of each method, particularly regarding the changes in the surface properties caused by the different preparation methods.
