Oxygen Gradient Measurement

Oxygen gradient measurement is the quantitative assessment of how dissolved oxygen concentration changes across space or within a biological system, revealing conditions that influence cellular function and microbial activity. The measurement typically uses oxygen-sensitive probes, microsensors, or imaging indicators to detect local oxygen levels, while diffusion, transport, and cellular respiration produce concentration differences between oxygen-rich and oxygen-depleted regions. In biology, mapping these gradients helps characterize tissue microenvironments, biofilms, organoids, and cell cultures, where oxygen availability can regulate metabolism, growth, signaling, and survival. These measurements support studies of development, disease, host-microbe interactions, and the design of controlled experimental systems.

This paper reports a polydimethylsiloxane-polycarbonate (PDMS-PC) microfluidic device capable of reliably generating combinations of chemical and oxygen gradients for cell migration studies, which can be practically utilized in biological labs without sophisticated instrumentation.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

Characterization of Sickling During Controlled Automated Deoxygenation with Oxygen Gradient Ektacytometry

Here, we present oxygen gradient ektacytometry, a rapid and reproducible method to measure red blood cell deformability in samples from patients with sickle cell disease under controlled deoxygenation and reoxygenation. This technique provides a way to study red blood cell sickling and to monitor sickle cell disease treatment efficacy.
