Pericyte Astrocyte Co-culture

Pericyte-astrocyte co-culture is an in vitro model that grows vascular pericytes and neural astrocytes together to study their interactions within the neurovascular unit. In shared or compartmentalized culture systems, the cells communicate through direct contact and soluble factors, allowing researchers to examine how astrocytes influence pericyte behavior and how pericytes affect vascular stability, barrier properties, and inflammatory signaling. This model supports investigations of blood-brain barrier regulation, neurovascular function, and disease-related changes, including responses to injury or inflammation. It also provides a controlled platform for testing cellular mechanisms and evaluating potential therapeutic strategies before experiments in animal models.

Astrocytes have been recognized to be versatile cells participating in fundamental biological processes that are essential for normal brain development and function, and central nervous system repair. Here we present a rapid procedure to obtain pure mouse astrocyte cultures to study the biology of this major class of central nervous system cells.

(2019)This video demonstrates the visualization of intracellular cargo transport in cultured astrocytes using a fluorescent acidotropic probe that selectively labels acidic endolysosomal vesicles. Time-lapse imaging and Z-stack acquisition with a confocal microscope allow tracking of fluorescently labeled cargo throughout the three-dimensional cell volume.

A Simple Technique to Isolate and Culture Murine Astrocytes

The video demonstrates a simple method to culture murine astrocytes in isolation from other brain cells. During culturing, by using astrocyte-specific media and inhibiting the adherence of non-astrocytic cells, the method selectively cultures astrocytes. These cultured astrocytes exhibit stellate morphology with fine processes.
