White Preadipocytes

White preadipocytes are precursor cells that develop into white adipocytes, the specialized cells that store energy in lipid droplets and contribute to endocrine regulation of metabolism. Their differentiation, known as adipogenesis, is initiated by signals such as insulin and glucocorticoids that activate transcription factors including PPARγ and C/EBP, driving changes in gene expression, lipid accumulation, and mature adipocyte function. In biology research, cultured white preadipocytes provide a controlled model for studying fat-cell development, adipose tissue formation, obesity, and insulin resistance, while also supporting the evaluation of compounds that alter adipogenesis or metabolic activity.

Isolation and Differentiation of Primary White and Brown Preadipocytes from Newborn Mice

This report describes a protocol for the simultaneous isolation of primary brown and white preadipocytes from newborn mice. Isolated cells can be grown in culture and induced to differentiate into fully mature white and brown adipocytes. The method enables genetic, molecular, and functional characterization of primary fat cells in culture.

Exogenous mitochondria of human and murine origin can be internalized by differentiating preadipocytes and tracked throughout adipogenic differentiation using imaging and genetic tools. While these mitochondria physically interact with the endogenous mitochondrial network, they do not increase the expression of markers associated with mature brown adipocytes.

Isolation of Preadipocytes from Broiler Chick Embryos

The present protocol describes a simple method for isolating preadipocytes from adipose tissue in broiler embryos. This method enables isolation with high yield, primary culture, and adipogenic differentiation of preadipocytes. Oil Red O staining and lipid/DNA stain measured the adipogenic ability of isolated cells induced with differentiation media.

Here we present methodology for the production of a focal stroke in murine white matter by local injection of an irreversible endothelial nitric oxide synthase (eNOS) inhibitor (L-Nio). Presented are two stereotactic variations, retrograde neuronal tracing, and fresh tissue labeling and dissection that expand the potential applications of this technique.
