Zebrafish Disease Model

A zebrafish disease model uses the vertebrate species Danio rerio to reproduce, investigate, or monitor features of human disease in a living organism. Because zebrafish embryos develop externally, remain transparent, and share many conserved genes and biological pathways with humans, researchers can introduce genetic changes or expose animals to candidate compounds and observe disease-related phenotypes. These models support studies of development, infection, cancer, cardiovascular disorders, and drug responses through imaging, behavioral analysis, and genetic screening. Their rapid development, reproductive capacity, and relatively low maintenance requirements make zebrafish valuable for studying disease mechanisms and prioritizing treatments for further investigation.

Renal injuries incurred from nephrotoxins, which include drugs ranging from antibiotics to chemotherapeutics, can result in complex disorders whose pathogenesis remains incompletely understood. This protocol demonstrates how zebrafish can be used for disease modeling of these conditions, which can be applied to the identification of renoprotective measures.

2026

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Zebrafish (Danio rerio) are widely used as an alternative vertebrate model organism in biomedical research. Their genetic similarity to humans, rapid development, and suitability for large-scale studies make them valuable tools...

Examining Muscle Regeneration in Zebrafish Models of Muscle Disease

2021

Skeletal muscle regeneration is driven by tissue resident muscle stem cells, which are impaired in many muscle diseases such as muscular dystrophy, and this results in the inability of muscle to regenerate. Here, we describe a protocol that allows the examination of muscle regeneration in zebrafish models of muscle disease.

A Possible Zebrafish Model of Polycystic Kidney Disease: Knockdown of wnt5a Causes Cysts in Zebrafish Kidneys

We describe a method of generating a possible zebrafish model of polycystic kidney disease. We used Tg(wt1b:GFP) fish to visualize kidney structure. Knockdown of wnt5a was by morpholino injection. Pronephric cyst formation after wnt5a knockdown was observed in this GFP transgenic zebrafish.
