His6 Tag Incorporation

His6 tag incorporation is a molecular biology technique that adds a six-histidine sequence to a recombinant protein, enabling its purification and detection. The histidine residues bind immobilized nickel or cobalt ions through coordination chemistry, allowing tagged proteins to be captured by immobilized metal affinity chromatography and released with imidazole or altered pH. In immunology and infection research, His6-tagged antigens, antibodies, and pathogen-associated proteins support protein production, antigenicity studies, assay development, and structural or functional analysis. This approach can streamline the preparation of defined research reagents, although tag placement and purification conditions may influence protein folding, activity, or accessibility.

Here, we present a protocol to generate a proof-of-principle divalent adenovirus type 5 (Ad5) vector Ad5/H5-HVR1-KWAS-HVR5-His6 by utilizing the Antigen Capsid-Incorporation strategy. This vector was demonstrated to exhibit qualitative fitness, the capability to escape Ad5-positive sera in vitro, and the antigenicity as well as immunogenicity to the incorporated antigens.
