Dna Purity

DNA purity is a measure of how free an extracted DNA sample is from contaminants such as proteins, phenol, salts, and RNA, and it is distinct from DNA integrity. Purity is commonly assessed by spectrophotometry, using absorbance ratios such as A260/A280 and A260/A230, while gel electrophoresis can reveal degradation or residual impurities that measurements may miss. In medicine and biomedical research, high-quality DNA supports reliable PCR, sequencing, genotyping, and molecular diagnostics because contaminants can inhibit enzymes, distort measurements, or reduce assay sensitivity. Evaluating purity alongside concentration and integrity improves the consistency and clinical relevance of DNA-based analyses.

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

Here we describe a robust method of determining immune cell identity and purity through epigenetic signatures detected using quantitative PCR (qPCR). DNA demethylation at a specific locus serves as a unique identifier for a particular cell type and allows for identification of CD8+, regulatory, or Th17 T cells.

Assessment of DNA Contamination in RNA Samples Based on Ribosomal DNA

Here, we present a protocol for tracing genomic DNA (gDNA) contamination in RNA samples. The presented method utilizes primers specific for the internal transcribed spacer region (ITS) of ribosomal DNA (rDNA) genes. The method is suited for reliable and sensitive detection of DNA contamination in most eukaryotes and prokaryotes.
