Mucus Secreting Goblet Cells

Mucus-secreting goblet cells are specialized epithelial cells that produce and release mucus to protect and lubricate internal body surfaces. They synthesize mucins, large glycoproteins, in the endoplasmic reticulum and Golgi apparatus, package them into secretory vesicles, and release them by exocytosis; contact with water then expands the mucins into a hydrated gel. Found in tissues such as the respiratory and gastrointestinal tracts, goblet cells help trap particles and pathogens, reduce friction, and support epithelial defense. Changes in their number or activity can influence airway inflammation, intestinal diseases, and responses to environmental or infectious stress.

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.

Inter-cellular communication is critical for controlling various physiological activities within and outside the cell. This paper describes a protocol for measuring the spatio-temporal nature of single cell secretions. To achieve this, a multidisciplinary approach is used which integrates label-free nanoplasmonic sensing with live cell imaging.
