Murine Colon Cells

Murine colon cells are cells derived from the mouse large intestine, including epithelial, immune, stromal, and neural populations that together support barrier function, host defense, and intestinal homeostasis. In vivo, epithelial cells renew from stem-cell compartments, while tight junctions and mucus-producing goblet cells regulate passage of luminal contents and limit microbial contact with underlying tissue. In biology research, murine colon cells are examined in tissue sections, primary cultures, organoids, and genetically modified mouse models to study epithelial repair, inflammation, host-microbe interactions, colorectal disease, and responses to candidate therapies. These systems connect cellular mechanisms with organism-level physiology, although findings require careful comparison with human colon biology.

The purpose of this protocol is to imitate human group B Streptococcus (GBS) vaginal colonization in a murine model. This method may be used to investigate host immune responses and bacterial factors contributing to GBS vaginal persistence, as well as to test therapeutic strategies.

A reliable method for the RNA isolation of Pseudomonas aeruginosa recovered from murine cecums is described. The RNA recovered is of sufficient quantity and quality for subsequent qPCR, transcription profiling, and RNA Seq experiments. This technique can be adapted for RNA isolation of other intestinal microbes.

In this video, we demonstrate the isolation of lamina propria mononuclear cells from murine colonic tissue by enzymatic digestion using collagenase E and silica-based density separation media gradient separation. The isolated mononuclear cells can be used for subsequent downstream analysis.
