Tsa-201 Cells

Tsa-201 cells are a human embryonic kidney cell line engineered to express the simian virus 40 (SV40) large T antigen, making them a widely used system for producing recombinant proteins in cultured cells. The antigen supports replication of plasmids containing an SV40 origin and promotes high-level, transient gene expression after transfection, while the cells remain amenable to routine laboratory culture. In biology and biomedical research, Tsa-201 cells are especially valuable for heterologous expression and functional analysis of ion channels, receptors, and other membrane proteins. They support electrophysiology, fluorescence imaging, and biochemical assays that link protein expression to cellular function.

Visualization of Mitochondrial DNA Replication in Individual Cells by EdU Signal Amplification

We developed a sensitive technique to label newly synthesized mitochondrial DNA (mtDNA) in individual cells in order to study mtDNA biogenesis. The technique combines the incorporation of EdU together with a tyramide signal amplification (TSA) protocol to visualize mtDNA replication within subcellular compartments of neurons.

Assessing Phagocytic Clearance of Cell Death in Experimental Stroke by Ligatable Fluorescent Probes

We present a new fluorescence technique for selective in situ labeling of active phagocytic cells, which clear off cell corpses in stroke. The approach is important for assessing brain reaction to ischemia because only a small proportion of phagocytes present in ischemic brain participate in clearance of cell death.

Human Neuroendocrine Tumor Cell Lines as a Three-Dimensional Model for the Study of Human Neuroendocrine Tumor Therapy

We present a simple agarose overlay platform to grow 3D multicellular spheroids using neuroendocrine cancer cell lines. This method provides a very convenient way to examine the effect of therapeutic drugs on the neuroendocrine tumor cells. It could also help us establish human neuroendocrine tumor spheroids for cancer therapy.

We demonstrate here that epigenetic reprogramming via Somatic Cell Nuclear Transfer (SCNT) can be used as a tool to generate mouse models with pre-defined T cell receptor (TCR) specificities. These transnuclear mice express the corresponding TCR from their endogenous locus under the control of the endogenous promoter.
