Cecal Slurry Preparation

Cecal slurry preparation is a laboratory method for converting cecal contents into a defined suspension for experimental studies of polymicrobial infection. The process involves collecting cecal material, homogenizing it in a sterile diluent, and standardizing the resulting slurry so it can be administered consistently across experimental groups. In immunology and infection research, this inoculum models exposure to a complex microbial community and enables investigators to examine host inflammatory responses, bacterial dissemination, immune regulation, and disease severity under controlled conditions. Careful preparation improves reproducibility when evaluating infection mechanisms, therapeutic interventions, and outcomes in animal models.

Experimental sepsis can be induced in mice using the cecal ligation and puncture (CLP) method. Current protocols to assess autophagy in vivo in the context of CLP-induced sepsis are presented here: A protocol for measuring autophagy using (GFP)-LC3 mice, and a protocol for measuring autophagosome formation by electron microscopy.

This protocol describes techniques to measure infectious outcomes underlying secondary hospital-acquired infections in the immunosuppressive condition, first by establishing cecal ligation/puncture mice then challenging them with intranasal infection to create a clinically relevant model of immunosuppression sepsis.

This protocol presents the operative details of cecal ligation and puncture (CLP) in a mouse model of sepsis. CLP is one of the most widely used techniques to create an animal model of sepsis. Therefore, a standardized CLP protocol is required for the attainment of reliable research results.
