Pseudo Pronuclei Analysis

Pseudo pronuclei analysis is a microscopy-based approach for identifying and characterizing pronucleus-like nuclear structures, helping researchers assess abnormal or experimentally induced nuclear remodeling. These structures arise when decondensed genetic material becomes enclosed by nuclear membranes without following the complete sequence of conventional fertilization, so analysis typically examines their number, size, morphology, and timing of formation. In immunology and infection research, the method can support studies of cell fusion, pathogen-associated changes in nuclear organization, and experimental conditions that disrupt genome handling. Quantitative imaging provides measurable endpoints for comparing treatments, clarifying mechanisms, and evaluating cellular responses.

Designer nucleases such as zinc finger nucleases (ZFNs) and transcription activator-like effector nucleases (TALENs) can be used to modify the genome of mouse preimplantation embryos by triggering both the nonhomologous end joining (NHEJ) and homologous recombination (HR) pathways. We demonstrate here that epigenetic reprogramming via Somatic Cell Nuclear Transfer (SCNT) can be used as a tool to generate mouse models with pre-defined T cell receptor (TCR) specificities. These transnuclear mice express the corresponding TCR from their endogenous locus under the control of the endogenous promoter.

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Salmonella invades and replicates inside intestinal epithelial cells both in Salmonella-specific vacuoles and free in the cytosol (hyper-replication). A high-throughput fluorescence microscopy-based protocol is described here to quantify the intracellular phenotypes of Salmonella by two complementary image analyses through ImageJ, reaching single-cell resolution and scoring.
