Thymocyte Apoptosis

Thymocyte apoptosis is the programmed death of developing T cells in the thymus, a process essential for shaping a functional and self-tolerant immune repertoire. During thymic selection, thymocytes that fail to recognize major histocompatibility complex molecules or bind self-antigens too strongly receive death signals that activate pro-apoptotic BCL-2 family proteins, mitochondrial cytochrome c release, and downstream caspases. This selective elimination supports central tolerance by preventing potentially autoreactive T cells from entering the circulation. In immunology and infection research, studying thymocyte apoptosis helps explain immune development, pathogen-associated changes in thymic function, immunodeficiency, and mechanisms that may contribute to autoimmune disease.

In this video, we describe a method to identify the small-molecule kinase inhibitors that modulate the apoptosis of self-reactive CD4+CD8+ double-positive immature thymocytes. Apoptosis is induced in the double-positive thymocytes by activating them with anti-CD3- and anti-CD28-coated magnetic beads; this is followed by a small-molecule inhibitor treatment and flow cytometry analysis to detect if the inhibitors modulate the apoptotic marker expression.

Activation of Apoptosis by Cytoplasmic Microinjection of Cytochrome c

In this protocol, we describe the direct cytoplasmic microinjection of cytochrome c protein into fibroblasts and primary sympathetic neurons. This technique allows for the introduction of cytochrome c protein into the cytoplasm of cells and mimics the release of cytochrome c from mitochondria, which occurs during apoptosis.

Apoptosis is a combination of two Greek words, 'apo' and 'ptosis,' meaning separation and falling off, respectively. Hippocrates used this word to describe gangrene, which was caused due to bandaging of fractured bones. Apoptosis was distinguished from necrosis in 1970 when John Kerr reported observations of morphological changes occurring during apoptosis. During one experiment, he observed that the disruption of blood supply to the liver tissue resulted in a size reduction of the tissue.

Early detection of apoptosis may identify at-risk cell populations in a variety of diseases. Here we demonstrate a method to link an early apoptosis-detection protein (Annexin V) to a MRI-detectable iron oxide nanoparticle (SPIO). This method may be extended to other proteins of interest to generate MRI-detectable molecular imaging probes.
