Amyloid Beta Secreting Cells

Amyloid beta secreting cells are cells that produce and release amyloid beta (Aβ) peptides, providing an important model for studying neuronal function and Alzheimer’s disease biology. Aβ arises when amyloid precursor protein is cleaved first by beta-secretase and then by gamma-secretase, generating peptides that can be secreted into the extracellular environment and, under some conditions, form aggregates. In neuroscience research, these cells help investigators examine amyloid production, peptide accumulation, cellular toxicity, and interactions with surrounding neural cells. They also support evaluation of disease mechanisms, biomarkers, and experimental treatments designed to reduce Aβ generation or limit its harmful effects.

Transcellular protein interactions are important determinants of pancreatic beta-cell function. Detailed here is a method—adapted from a coculture model of synaptogenesis—for investigating how specific transmembrane proteins influence insulin secretion. Transfected HEK293 cells express proteins of interest; beta cells do not need to be transfected or otherwise directly perturbed.

This protocol allows one to identify factors that modulate functional beta cell mass to find potential therapeutic targets for the treatment of diabetes. However, using live-cell confocal microscopic imaging techniques with fluorescent dyes, the type and kinetics of cell death as well as the pathways utilized can be studied in greater detail.
